Control of Golgi-enzyme sorting in development and disease

Protein glycosylation is known to be essential for tissue integrity and human development. It is therefore not surprising that devastating illnesses such as developmental abnormalities, different forms of cancer and inflammation are associated with alterations in glycosylation. Despite their known importance, the processes controlling the structures of different oligosaccharides (aka glycans) are poorly understood. Our interest is in the mechanism and control of intra-Golgi protein sorting that determines localisation of the processing enzymes and thereby glycan structures. We will be using biochemical, cellular and whole animal approaches to study membrane trafficking. These will be combined with modern imaging and mass spectrometry based glycobiological methods to uncover the organisation and control of the processing machinery in the Golgi. Our results will help to develop new diagnostics and therapies for developmental disorders in glycosylation. We will also contribute to the understanding and potential treatment of wide spread diseases such as cancers or chronic inflammation in which glycosylation changes are well documented during pathology.
