The purpose of the research proposed here is to find binding partners of the oncogenic DNA-binding complex composed of proteins LMO2-LDB1-SCL/TAL1-E47 and to define how, at a molecular level, these interactions might be important for haematopoietic development and malignancies. To map the details of these interactions, we plan to use a variety of techniques such as co-immunoprecipitation, immunodepletion experiments, pull-down assays. Further, we plan to characterise these complexes at a structural level by using X-ray crystallography of key binding components.with the haemopoietic specific LMO2-Ldb1-SCL/Tal1-E47 complex.
