FULLY FUNDED PhD: Studying drug resistance in biliary cancer through the investigation of the cross-talk between cancer and micro-environment

Project Description

Biliary cancer are highly chemo-resistant. Drug resistance is the result of intrinsic properties of cancer cells, as well as of the cross talk between cancer and other cell types.

We know that:
1) biliary cancer stroma is characterised by an immune-suppressive milieu that affects chemo-sensitivity in resected disease,
2) circulating immunosuppressive cells (such as monocytes) hold a negative prognostic value in patients undergoing chemotherapy,
3) monocytes and macrophages promote biliary cancer progression in animal models,
4) cancer stem cells inform the behaviour of microenvironemental cells.

In this project we will use biliary cancer patients’ derived organoids as a model of biliary cancer derived from cancer stem cells to investigate drug resistance, cross-talk with immune cells and effect of immune cells onto drug resistance.

The candidate will develop skills in primary cell cultures, patients’ derived organoids establishment and culture, isolation of peripheral blood cells, nucleic acid extraction and processing for genomic and transcriptomic analyses, culture of monocytes-macrophages, cloning, extracellular vescicles isolation and analysis, protein assessments, co-cultures, assessment of drug sensitivity.
