The platelet is a blood cell that plays a key role in the prevention of bleeding. For platelets to form a clot or thrombus effectively, they must rearrange their actin cytoskeleton to withstand high shear stress from blood flow in the arteries. This process depends on the environment of the artery as platelets change behaviour in response to matrix type, concentration and stiffness. Intracellular cyclic nucleotide messengers cAMP and cGMP reverse platelet activation and alter the platelet actin cytoskeleton, causing thrombus instability under high shear stress. But exactly how this response is controlled by specific extracellular matrix characteristics is unknown. We hypothesise that the type, concentration and stiffness of specific matrix proteins alter platelet responses to cyclic nucleotide signalling to affect thrombus stability and structure. To test this hypothesis, you will use a combination of immunobiological approaches, imaging and functional assays to examine how matrix proteins modulate cyclic nucleotide signalling in human platelets. This will reveal new insights into how the platelet environment controls thrombus development. This is a full time PhD in Medical Sciences.
