About the Project
Many tasks within the cell are accomplished by highly organized multi-component protein complexes, the biogenesis of which requires carefully guided assembly by molecular chaperones. A particularly striking example is the molecular machinery that drives movement of motile cilia. Motile cilia are present on the surface of many types of cells, and their regular beating is required of mucociliary clearance in airways, male and female fertility, brain development, and development of left-right organ asymmetry. Primary Ciliary Dyskinesia (PCD) is a rare inherited disease (primarily of children) in which cilia to stop beating. PCD patients consequently have symptoms attributable to failure in these processes (recurrent lung infections, immotile sperm, defective Fallopian tubes, as well as symmetrical internal organs). Most of the >30 genes currently known to cause PCD when mutated encode either subunits of the dynein motor protein complexes that drive ciliary beating, or encode proteins required to build these motor complexes during cilium construction – they appear to be ’molecular chaperones’ that recruit HSP90. However, very little is known about their molecular function as chaperones.
In this project, the student will use state of the art imaging (both fluorescence and electron microscopy) to investigate the dynein assembly and transport processes. This will utilise Drosophila as a powerful model for a structural and functional analysis of motor complex assembly in vivo.
This 4 year PhD project is part of a competition funded by EASTBIO BBSRC Doctoral Training Partnership. This opportunity is open to UK and international students and provides funding to cover stipend and UK level tuition fees. The UoE covers the difference between home and international fees. There is a cap on the number of international students the DTP can recruit so it is important to know in which fees status category applicants fall under when applying.
