About the Project

This project integrates structural and cell biology to better understand and treat a major, but under-recognised, disease process. Approximately 45% of deaths in the developed world result from fibrosis (scarring) of vital organs: more people are living longer with chronic disease, and we lack highly effective drugs to treat fibrosis. The project will use biophysics (ITC), structural (X-ray crystallography, NMR spectroscopy), and cellular studies to improve our understanding of important pro-fibrotic mechanisms and help develop drugs to target them. Dr Campeotto has optimised methods to make and purify the important pro-fibrotic protein NOX4. Prof Gooptu’s group have recently identified a novel molecular pathway mediating epithelial responses to lung injury triggers that worsen (exacerbate) fibrosis. The project will focus upon NOX4 and 2 key exacerbation pathway components, all validated as attractive therapeutic targets in cells and in vivo. We will test the conclusions from structural studies in our cell models of disease.
