Project Description

Juvenile idiopathic arthritis (JIA) is the most prevalent childhood rheumatic disease and is characterised by inflammation of the joints and other tissues that persists for more than six weeks. JIA is a heterogeneous disease, with multiple subtypes that are characterised by different clinical features. To date, genome-wide association studies (GWAS) have identified 28 loci underlying disease susceptibility, but the lack of progress in determining the causal genes at these loci has limited the clinical translation of these genetic discoveries for the development of novel treatment options. The overall objective of the project will be to localise causal genes for JIA and understand the molecular mechanisms through which they impact on the disease. The project will make use of GWAS of JIA and other autoimmune conditions in the largest sample sizes available to the supervisory team (and through collaborators) to catalogue disease loci. Throughout, we will make use of existing statistical genetics methodology and novel state-of-the art techniques under development by the supervisory team to fine-map JIA GWAS loci and integrate these genetic data with a range of multi-omics resources. The project will provide the most comprehensive view of causal genes at JIA GWAS loci and the molecular mechanisms through which they impact on subtypes of the disease and other autoimmune conditions. Localisation of causal genes provides an opportunity to prioritise potential new targets for therapeutics and inform on treatments that might be relevant to specific JIA subtypes.
